1. Pre-freezing Stage: Vaccines pre-filled in vials are placed on the shelf of the drying chamber. The shelf temperature is rapidly reduced to -40℃ to -80℃ via a high-efficiency cascade refrigeration system, freezing the water in the vaccine into ice crystals. This step prevents the formation of large ice crystals that could damage the vaccine’s protein structure or nucleic acid chains.
2. Primary Drying Stage: The drying chamber is evacuated to a high vacuum level (≤1 Pa). The shelf temperature is gradually raised to sub-zero temperatures, triggering the direct sublimation of ice crystals into water vapor without passing through the liquid phase. The water vapor is captured by the ultra-low-temperature cold trap (≤-85℃), ensuring the vaccine maintains a porous, loose structure.
3. Secondary Drying Stage: The shelf temperature is further increased to 20℃ to 30℃ to remove the bound water adsorbed on the vaccine matrix. This step reduces the residual moisture content of the vaccine to ≤1%, effectively inhibiting microbial growth and protein denaturation, thus ensuring long-term storage stability.
